Okay, I think I'm to the point where I need some specifics, but feel free to put it in a spoiler tag!
At the point where I have control of two turrets, and have looooads of zombies infinitely running towards me at various intervals of time, I'm stuck staring at a portal. I recognize that I'm probably supposed to be tossing grenades somewhere, and have aimed at every point that looks promising on both the upper walkway and lower floor without any luck. So... help, please?

Wait... I did turn off the valve below!
But I'm not sure where the valve on the upper/walkway level is. I honestly have no idea.
edit. Ah, I see. I think I reloaded after the bottom valve, and thus never noticed the new valve (I KNEW it was there, too!). Thanks, guys!

So, I got to the walkway where I am being pursued by zombies, threw a grenade to blow open the door, ran through it and turned the wheel of the valve.
I can't turn it anymore and nothing new has appeared. What do I do now?

Go back upstairs once you've turned the valve and have a really careful look around. (if you're still stuck just say and someone will walk you through, but we reckon it's more fun to work it out for yourself....)

I'm stuck in Episode 1 in the room with the auxiliary generator node. Minerva just asked me to open the locked door with a combine lock on it. I can't work out how to do that. There's an infinite machine gun ammo crate nearby, and I've shot everything a lot, but that doesn't help.
